Biosynthesis of aminocyclitol antibiotics |

Aminocyclitol antibiotics have been used clinically since streptomycin and neomycin first appeared. Biosynthetic study on these antibiotics is very limited until now. We have cloned sequenced and characterized the biosynthetic genes if 2-deoxystreptamine containing antibiotics; gentamicin, kanamycin, and developed a method for cloning the biosynthetic genes of 2-deoxystreptamine (2-DOS) containing aminoglycosides. The two gene clusters are similar which are in accordance with the similarity of gentamicin and kanamycin structures. 2-deoxystreptamine (2-DOS) is formed by the action of 2-deoxy--scyllo-inososes synthase (2-DOIS), followed by 2-deoxy-scyllo-inosose aminotransferases and dehydrogenase (oxidoreductase). We have expressed 2-DOIS gene in the heterologous host (E. coli BL21 DE3) and verified its function in vitro. Similarly, the gene has been inactivated by the insertion of thiostrepton or neomycin resistance genes to proved its role in vivo. The manipulation of 2-DOS-biosynthetic genes has paved the way to develop hybrid aminoglycosides. |
