Oligonucleotide@Doxorubicin Nanoaggregates with Dual-responsive Linkers toward Intracellular Release |

The nanoaggregates co-encapsulating doxorubicin and oligonucleotide were prepared for dual-responsive breakdown of the nanostructure with complete disappearance characteristics. 4-arm-poly(ethyleneglycol) was co-conjugated with doxorubicin and anti-bcl2 oligonucleotide with reducible and pH sensitive linkers, respectively. The conjugate was self-assembled into nanoaggregate in aqueous solution. Elemental scanning of the nanoaggregates revealed the core-shell structure of the nanoaggregates. The tracking of size modulation suggested a complete disappearance of the particles at reducing conditions and the liberation of oligonucleotide at low pH. The released fractions were analyzed by reversed phase chromatography. The nanoaggregates showed the lowest IC50 value when a cytotoxicity assay was performed against A549 cells. Apoptosis assay also confirmed that cells treated with the nanoaggregates showed higher fluorescence intensity of anti-apoptotic antibody compared to native doxorubicin. |
