제목 3-Hydroxypropionic acid Tolerance in Klebsiella pneumoniae: The Role of Transcriptional Regulator YieP and the Acid Transporter YohJK.
초록 Acid tolerance is an important issue for industrial production of chemicals, such as 3-hydroxypropionic acid (3-HP). In Escherichia coli, the transcription factor, YieP, was reported to regulate yohJK, genes encoding putative 3-HP exporter(s). The deletion of YieP could greatly improve the 3-HP tolerance via overexpression of yohJK. In this study, similar phenomenon was observed in Klebsiella pneumoniae, an important host for 3-HP production.Protein comparison showed that YieP and two putative 3-HP transporters (YohJK1 and YohJK2) are present K. pneumoniae. The deletion of YieP improved 3-HP tolerance in K. pneumoniae. Moreover, transcription analysis shown that YieP negatively regulates YohJK1 but not YohJK2, suggesting that YohJK1 is the ‘true’ 3-HP exporter in K. pneumoniae. Overexpression of YohJK1 could improve 3-HP production in the 3-HP stressful condition.
