The investigation of the effects of micropatterns and cell-adhesive peptide on myoblast functions using enzyme-triggered hydrogels |
초록 |
Myoblast adhesion and formation of multinucleated myotube in anisotropic patterns are important steps during muscle regeneration processes. It is also well known that adhesion and growth of myoblasts are regulated by chemical and structural properties of underlying substrates. However, there has been no report on whether the influences of their factors are synergistic or independent to each other. Given that, we prepared peptide-incorporated hydrogels of (1) various peptide density (0.19 ± 0.04 to 0.89 ± 0.08 nmol/cm2) and (2) micro-scaled grooves with various widths (10, 25, 80 μm). When myoblasts were cultured on these hydrogels, we found that the adherent morphology, proliferation and differentiation were affected by peptide concentration and groove size, however, peptide concentration seems to be a dominant regulator on myoblast functions. |
