Horseradish peroxidase (HRP) is a calcium-dependent, heme-containing oxidoreductase found in a plant Armoracia rusticana. Due to its broad substrate specificity, colorimetric, fluorometric, chemiluminescent and electrochemical assays for the diagnostic purposes have been successfully devised using a wide variety of substrates. Obviously, further improvements in the application and/or stability of the enzyme have been sought. In this regard, the production of recombinant HRP in bacteria such as Escherichia coli can be a good alternative and meet the required demands. In our present study, efforts were made to express HRP in the soluble and active form by the fusion of a soluble tag called phosphoglycerate kinase (PGK) to its N-terminus and co-expression of ferrochelatase. |
