pH-dependent color assay for glycosyltransferase |

The major method for glycosyltransferase activity assay is TLC, HPLC and MS analysis. But there are so many troublesome especially about aminoglycoside related glycosyltransferases. Here we report simple and sure method for aminoglycoside glycosylation based on pH change. Cresol red is pH indicating compound the range from pH 6 to pH 8. The molecular structure of that is easily change depend pH exchange so that the color is yellow at low pH and red at high pH. The intensity alteration was detected at 550nm with and without glycosyltransferase. This pH-dependent method will be useful tool for HTP screening of glycosyltransferase mutagenesis. |
