Three-dimensional cell mass composed of fibroblast to enhance cell therapeutic efficacy |

Cell spheroid has received much attention from many researchers to overcome the typical limitations for cell-based therapy. In this study, we developed the 3D cell mass (3DCM) composed of fibroblast (FB) which is well known to produce the extracellular matrix (ECM) and cytokine and investigated its the wound healing efficiency. The 3DCM was prepared by regulation of the cell-integrin interaction. Briefly, FB was seeded to MBP-FGF fusion protein coated wellplate. After that, FB was aggregated to form 3DCM spontaneously. ELISA results of the FB 3DCM were demonstrated that higher amount of various angiogenic factors were secreted as compared to monolayer-cultured FB. Secreted ECM components from FB 3DCM were increased significantly. The in vivo evaluation of wound healing efficacy using nude mice was under investigated. Obtained results suggest that FB 3DCM formed by modulation of cell-integrin interaction is a promising candidate for cell therapy. |
