제목 Facile and Large-scale Fabrication of Exosome-analogous Nanovesicles by Extrusion of Cells with Novel Cell Membrane Detergent
초록 We propose an exosome-analogous drug delivery system which is established by cell extrusion process with aids of a newly synthesized cell membrane detergent, mannosylerythritol lipid-conjugated PEG (MEL-PEG). Introduction of MEL-PEG facilitates large-scale fabrication of exosome-mimetic nanovesicles by deforming plasma membrane without denaturation of biomolecules, and exposes PEG chains at the surface of generated nanovesicles. We analyzed enhanced production yield of nanovesicle and detected exosome marker proteins by western blot. Finally, we demonstrated increased circulation time of PEGylated exosome-mimetic nanovesicles. We expect that this facile generation system of exosome-analogous nanovesicles enables the practical applicability of exosome therapeutics.
