Optimization of One-Bead One-Compound (OBOC) Library Synthesis for Kinase Substrate Screening by Using MALDI-TOF MS |

We have developed the one-bead one-compound (OBOC) combinatorial library method for the screening of enzyme substrate specificity. In the OBOC library , each of the resin beads carries not only the full-length peptide product but also small amounts of sequence-specific N-terminated ones.. The sequence of the full-length peptide is determined by using MALDI-TOF MS, which measures the mass differences between the adjacent amino acid added peptides and the N-terminated peptides. We optimized the condition to control the amounts of N-terminated peptide products to give the maximum purity of the full-length peptide. |
