Amphiphilic block copolymers have received much attention for their applications to multifunctional drug carriers because they can be self-assembled to encapsulate hydrophobic drugs with a high efficiency and circulate in the blood for a prolonged period of time. In this work, we investigated polyglycerol-block-poly(Ɛ-caprolactone) (PG-b-PCL) nanoemulsions incorporating anti-cancer drugs (paclitaxel) and contrast agents (lipiodol), followed by conjugation with folates via simply esterification of boronic acid intermediate. The prepared nanoemulsions had a diameter of 150-180 nm with 19.2% degree of folate substitution, encapsulated 2.6 % of paclitaxel with 55% efficiency, showed cytotoxicity in HeLa cells with LD50 < 3 ng mL-1, and enhanced cellular uptake and CT imaging contrast. This study demonstrates that PG-b-PCL can be used to prepare multifunctional nanocarriers for drug delivery and bioimaging. |
