제목 Development of process to produce epothilones using Sorangium cellulosum
초록 First, we investigated ammonium removal by cation resin to improve epothilone titer produced by Sorangium cellulosum. About 83 % of ammonium was removed when 8 g/l Amberite FPC22 Na was added at the death phase. As the result, epothilone B production enhanced from 5.45 mg/l to 14.3 mg/l. Second, we studied on cell immobilized system using calcium alginate and polyurethane foam. Epothilone B production using calcium alginate was more effective than using polyurethane foam. Futhermore, with the 3 % (w/v) calcium alginate, the prolonged repeated batch culture (five batch, 24 days) was carried out. Total epothilone B production and mean productivity reached 58.83 mg/l and 2.42 mg/l•day, respectively.
