Release and cytokine production of BmpB from BmpB-loaded pH-sensitive and mucoadhesive thiolated Eudragit microspheres |

Swine dysentery (SD) is a contagious mucohaemorrhagic colitis of pigs that is caused by anaerobic intestinal spirochaete Brachyspira hyodysenteriae. Recently, the mice or pigs immunized with an Escherichia coli clone expressing recombinant BmpB generated antibodies recognizing the native BmpB of B. hyodysenteriae. In this study, BmpB used as a vaccine candidate for oral delivery was encapsulated into the pH-sensitive and mucoadhesive Eudragit microspheres. The loading efficiency of BmpB was 75% and the sizes of the microspheres ranged from 5-15 µm. Mucoadhesive experiment in vitro showed that the adherence of thiolated Eudragit microspheres in porcine intestine was higher than the Eudragit microspheres. BmpB release profile was compared at stomach pH 2.0 and intestinal pH 7.2 resulted in 22-32% release at pH 2.0 and 82% release at pH 7.2 within 24 h. In vitro production of cytokines in RAW264.7 from BmpB-loaded thiolated Eudragit microspheres will be reported. |
