Quartz crystal microbalance with dissipation monitoring (QCM-D) was used to investigate the effect of heparin on fibrin formation from fibrinogen solution onto hydrophobic polystyrene surface. The anticoagulant function of heparin has been well known. Many studies have suggested that heparin prevent fibrin formation by means of binding to antithrombin, while some literatures have described the binding of heparin to thrombin. In the present study the effect of heparin on fibrin formation was studied quantitatively as a function of added heparin concentration in the absence of antithrombin as well as in the presence of antithrombin. Herein, QCM-D technique allows simultaneous measurements of the mass change and the viscoelastic property of deposited layers. Atomic force microscopy (AFM) was used to characterize the final morphology of polymerized fibrin layers. |
