제목 Tissue Engineered Spinal Cord
초록 We tested the hypothesis that transplantation of bone marrow stromal stem cells (BMSCs) into the spinal cord after a severe cord injury promotes functional outcome. Porous PLGA nerve guidance channels slowly releasing BDNF were prepared by means of ice particle leaching methods. Fischer rats were anesthetized and laminectomy was performed at the T8~T9 level. A full thickness transection of the spinal cord was made at 2 levels, and 5 mm length transected segment was removed. No treatment was done in animals (Group 1). Only guidance channels were transplanted into the gap in animals (Group 2). BMSCs seeded with nerve guidance channels were transplanted into the gap in animals (Group 3). Motor functional outcome measurements using BBB score were performed weekly to 8 wks post-injury. Sections of spinal cord tissue area were analyzed by H&E and IHC staining for NF, S-100 and NSE. The data indicate significant improvement in functional outcome in animals treated with BMSCs transplantation and BDNF compared to without BMSCs and BDNF. In histological evaluation evidence of axonal regeneration into the newly formed tissue at the gap was found in animals treated with BMSCs transplantation (Group 3), but very weak or no evidence of axonal regeneration was found in Groups 1 and 2 animals.
