Burkholderia glumae BGR1 produce 4-hydroxybenzoate (4HBA) as intermediate during p-coumaric acid (pCA) degradation at high speed. produced 4HBA from pCA is further metabolized through the protocatechuate 4,5-cleavage pathway. To accumulation of 4HBA in this reaction 4-hydroxybenzoate-3-monooxygenase (4HBMO, GI: 238023559) deletion mutant of B.glumae BGR1 was constructed and evaluate, which is characterized as involving in the ring cleavage. There is proposed β oxidation pathway of bioconversion of pCA to 4HBA in this strain.p-coumaroyl-CoA is formed by an ATP-, Mg2+, and CoA dependent feruloyl CoA synthase (FCS, GI: 238024454) reaction.
