Pyropheophorbide a-embedded Polymeric Nanophotosensitizer for in vivo photodynamic therapy of cancer |

Pyropheophorbide a (PPa) is a well-known photosensitizer for photodynamic therapy (PDT) of cancer. Use of PPa is often limited by poor colloidal stability under physiological condition and targeting efficiency to cancerous site. Here, we report PPa-embedded polymer nanoparticles (PPa NPs) with improved colloidal stability and cancerous cell/tissue accumulation. PPa NPs with the average size of ~20 nm were fabricated through repeated microemulsion in aqueous solution of a non-ionic surfactant (Pluronic F-68). The photo-induced therapeutic efficacy of PPa NPs was investigated in a SCC-7 tumor-bearing mouse model. We demonstrate that the PPa NP can efficiently induce tumor suppression by apoptosis or necrosis of cancerous cells by PDT treatment. |
