Dendrimer has synthetic barriers such as difficult purification and multi-step synthesis. We prepared the novel aliphatic ester dendrimer having the growing units of 2, 2-bis(hydroxymethyl) propionic acid. Polystyrene, as a support, was polymerized from benzyl hydroxyl derivative using ATRP. From one end of polystyrene support, dendrimer consisting of esterified bis-MPA units was grown up and purified by precipitation based on the poor solubility of polystyrene support. In the last step, we selectively decomposed the benzyl ester group between dendrimers and the support using Pd(Ⅱ) Acetate / H2 and obtained free dendrimers separated from polystyrene using the differences in solubility of each compound. The obtained dendrimer having ester-backbone is water-soluble, biodegradable and biocompatible. The simplified purification step in the proposed synthesis is attractive for the preparation of high-generation dendrimers in a large scale. |
