제목 A novel approach to the synthesis of aliphatic polyester dendrimers
초록 Dendrimer is defined as regular, highly branched and globular macromolecule with many tree-like arms emanating from a central core. Dendrimer has actual and potential advantages enable to control the size and high surface functionality. Due to the synthetic barriers such as difficult purification and multi-step synthesis, a reasonable cost and a coverage of use have been limited. Therefore, dendrimer is usually used to the areas of high value such as biomedical fields.
Hült group presented aliphatic dendritic polyesters based on 2, 2-bis(hydroxymethyl) propionic acid. Hydroxyl-terminated dendrimers up to the fourth generation require many chromatographic purification steps so that this method places a limit on the large scale dendrimer preparation. To overcome this problem, Fréchet group reported fast and convenient divergent synthesis of aliphatic ester dendritic hybrids up to the fourth generation without any chromatographic purification; however, it is not a dendrimer itself but a dendrimer conjugated with linear polymer as a dendritic hybrid.
We prepared the novel aliphatic ester dendrimer having the growing units of 2, 2-bis(hydroxymethyl) propionic acid. Polystyrene, as a support, was polymerized from benzyl hydroxyl derivative using ATRP. From one end of polystyrene support, dendrimer consisting of esterified bis-MPA units was grown up and purified by precipitation based on the poor solubility of polystyrene support. In the last step, we selectively decomposed the benzyl ester group between dendrimers and the support using Pd(Ⅱ) Acetate / H2 and obtained free dendrimers separated from polystyrene using the differences in solubility of each compound. The obtained dendrimer having ester-backbone is water-soluble, biodegradable and biocompatible. The simplified purification step in the proposed synthesis is attractive for the preparation of high-generation dendrimers in a large scale.
