Induction of osteogenic differentiation using the BMP-2 linked hydrogel and human periodontal ligament stem cells (hPDLSCs) |

Recently, bone morphogenetic proteins (BMP), especially BMP-2 and BMP-7, are mainly used for bone regeneration. However, the dosage of BMP for treatments is even higher than physiological concentration because of short half-life and instability of the BMP. In this work, we synthesized the MPEG-b-(ε-caprolactone–ran-α-chloro-ε-caprolactone) (MPEG-PCL-Cl), and then chlorides of the MPEG-PCL-Cl are substituted to azides. The BMP-2 containing alkyne group was chemically linked MPEG-PCL-Cl via click chemistry. We mixed this solution with hPDLSCs and implanted to dorsum of nude mouse by subcutaneous injection. Injected the solution immediately became a gel-state. hPDLSCs were proliferated and differentiated to osteoblastic cells. We monitored osteoblastic phenotypes and gene expression through histological methods and qRT-PCR respectively. In conclusion, We confirmed that BMP-2 linked MPEG-PCL-Cl hydrogels could increase a half life and a stability of the BMP-2. |
