Polydiacetylene liposomes have unique properties that allow liposomes to change color from blue to red and emit fluorescence in response to external stimuli such as temperature, pH, mechanical stress, organic solvent and antibody–antigen interaction. In this study, we demonstrate an antibody-based biosensor using PDA liposomes for detection of allergy diagnosis by hIgE. In order to detect target hIgE, the surface of immoblized PDA liposomes were functionalized with hIgE monoclonal antibodies and the fluorescent signals were detected after antigen-antibody immunoresponse. However, fluorescent signals were slightly increased depending on the target protein concentration when polyclonal antibody conjugated with HRP were used as an amplifying agent after primary immunoresponse. After secondary immunoresponse, HRP-biocatalyzed oxdiation of DAB in the presence of H2O2 formed precipitation of an insoluble product. |
