제목 Poly(2-dimethylaminoethyl methacrylate)-functionalized GO nanostructures for efficient cellular delivery of siRNA
초록 Despite the high therapeutic potential, the use of siRNA has still been limited due to the lack of an appropriate delivery system. In this study, we synthesized two-dimensional carriers of siRNA using a surface functionalized and easy method of dopa-catechol derived poly(2-dimethylaminoethyl methacrylate)(CD-poly(DMAEMA)) and graphene oxide(GO). The resulting GO/CD-poly (DMAEMA) can interact with siRNA on the flexible 2D surface of GO. It can wrap siRNA by folding to form multi-layered nanostructures, which provided excellent protection from the attack of nucleases. GO/CD-poly(DMAEMA) demonstrated significantly improved cellular uptake and enhanced gene silencing efficiency in the presence of serum, compared to bPEI 25kDa and unmodified GO. Considering its desirable transfection efficiency and low toxicity, GO/CD-poly(DMAEMA) would be potential candidate for siRNA delivery.
