Docetaxel (DTX) is currently available for treatment of various cancers in a clinic. However, its therapeutic effect has been severely limited by low solubility and indiscriminate toxicity to normal cells as well as cancer cells. In order to surmount these drawbacks, dextran derivatives have been extensively investigated as the drug vehicles because of their excellent water solubility and biocompatibility. Particularly, carboxymethyl dextran (CMD) has been often used to prepare polymer-drug conjugates, primarily owing to their multiple functional groups which allow for conjugations. In this study, we prepared CMD-based polymeric conjugates by a facile chemistry, which could improve the solubility of DTX and selectively release it in the mild acidic condition. The physicochemical characteristics of the CMD conjugate was investigated using 1H-NMR, DLS, HPLC, and in vivo imaging system. Overall, the conjugates could be useful as a drug carriers for improved cancer therapy. |
