Hypoxia-triggered drug deliveryusing carboxymethyl dextran-based self-assembled nanoparticles |

In this study, we attempted to prepare hypoxia-responsive nanoparticles (HR-NPs) that facilitate rapid release of drugs at hypoxic tumor tissue, resulting in enhanced therapeutic efficacy. To prepare hypoxia-responsive amphiphilic conjugates, a hydrophobic 2-nitroimidazole (NI) derivative was conjugated to the backbone of hydrophilic carboxymethyl-dextran through amide formation. Doxorubicin was encapsulated into the hydrophobic core of HR-NPs by the emulsion method. Due to the high sensitivity to hypoxia of NI derivative, the drug release rate of HR-NPs remarkably increased under the hypoxic condition. In addition, HR-NPs were selectively accumulated at the hypoxic tumor tissues after their systemic administration into the tumor-bearing mice. Overall, HR-NPs could be promising drug carriers for selective delivery of drugs into hypoxic cells. |
