제목 High throughput screening method for engineered lysine decarboxylase from E. coli strain B
초록 Cardaverine can be used as a monomer for synthesizing polyamide with dicarboxylate such as an adipic acid. In these days, depletion of petroleum resources and sustainable development are major issues, also in chemical industry. In such a viewpoint, production of polymers using cardaverine, which can be produced from L-lysine, is highly preferred. In this study, we set up a high throughput screening method for engineered lysine decarboxylase. We made a replica of engineered lysine decarboxylases library. Using a bromothymol blue as a pH indicator, we could screen a blue colony directly on the plate. As a result, we found a mutant which shows a higher activity in the α-decarboxylation reaction of L-lysine than wild type lysine decarboxylase.
