Preparation and release behavior of Ipriflavone-loaded PLGA scaffold for tissue engineered bone |

The regeneration of bone tissue is an ambitious goal in orthopaedic surgery for healing large bone defects secondary to tumor or trauma, and for treating delayed unions or non-unions. Recently, bone tissue engineering has become a possible alternative strategy to regenerate bone. In this study, we developed the novel IP loaded poly(L-lactide-co-glycolide)(PLGA) scaffolds for the application of the tissue engineered bone. IP/PLGA scaffolds were prepared by solvent casting/ice leaching method and were characterized by porosimeter, SEM. The release profiles of Ipriflavone were examined using pH 7.4 PBS by HPLC (for 84 days at 37 °C). Cellular viability and Osteogenic differential were assayed by CCK-8, ALP activity. The release of IP in vitro was more prolonged over 80 days. Formation of bone tissue was better IP/PLGA hybrid scaffold than control. As a result of this study, It seems that ipriflavone plays an important role for bone induction in IP/PLGA scaffolds. |
