제목 Culture of nucleus-pulposus cell in PLGA scaffold modified with SIS sponge in vitro
초록 Small intestinal submucosa(SIS) is a source of collagenous tissue with biocompatibility. In this study, we enhanced nucleus pulposus(NP) cell growth using SIS to complement lacked bioactivity of PLGA. Prepared PLGA scaffold was mixed SIS gel and cross-linked by EDC. Characterization of scaffold were measured. Scaffold seeded NP cell was cultured for several weeks. We confirmed DNA and collagen quantity in scaffold. RT-PCR was performed on type II collagen and aggrecan. Cell viability was evaluated by MTT assay. In PLGA modified with SIS sponge cell viability was high at 6 weeks than other scaffold. Gene expression of disc cell was good at SIS sponge at 1, 2 weeks, but good result showed at PLGA/SIS sponge in process of time. Also, GAG and collagen content presented similar result to RT-PCR. We suggests that PLGA scaffold modified with SIS sponge is suitable to form NP tissue. Acknowledgment : This work was supported by KMOHW (0405-BO01-0204-0006).
