제목 | Effect of SIS Scaffold on Chondrogenic Differentiation for Bone Marrow-derived Stem Cells |
초록 | Objective: Bone marrow-derived stem cells (BMSCs) may represent a convenient source of stem cells for cell therapy and tissue engineering application. They exhibit both multipotentiality and self-renewal capabilities. Small intestinal submucosa (SIS) had been widely used as a biomaterial without immune rejection responses. In this study, we manufactured that SIS sponge prepared by crosslinking with EDC and examined into osteogenic differentiation of BMSCs into SIS sponges. Methods: Fabricated SIS sponge was cross-linking by 100 mM EDC solution. Rat-BMSCs seeded 1x106cells in sponge and cultured in vitro for 28days. We have characterized the chondrogenic potential of rat-BMSCs in SIS sponges by histology, GAG, MTT assay, SEM and RT-PCR for chondrogenic specific phenotypes. Results: In SEM, all morphology of SIS sponges showed regular and interconnected pore structure. We know that BMSCs cultured with chondrogenic media displayed significantly higher levels of GAG content compared to untreated controls. By RT-PCR analysis, we observed type II collagen (chondrocyte specific genes) expression. This result demonstrates chondrogenic differentiation of rat-BMSCs. This result indicates that SIS sponge is useful for chondrogenesis of BMSCs. Conclusion: We confirmed that SIS sponge were highly porous with good interconnections between each pore, which can support the cell adhesion, proliferation, and differentiation.
