Solid free-form fabrication (SFF) of tissue engineered scaffold was successfully carried out with poly(lactic-co-glycolic acid) grafted hyaluronic acid (HA-PLGA) / polyethyleneglycol (PEG) blend system encapsulating intact BMP-2. PEG was blended with HA-PLGA to encapsulate BMP-2 without denaturation, which were exploited for the SFF to prepare tissue engineered scaffolds. In vitro release tests confirmed the sustained release of intact BMP-2 from the scaffolds up to a month. After the assessment of the proliferation, DNA content, and ALP activity of osteoblast cells, the blended HA-PLGA/PEG/BMP-2 scaffolds were implanted into the calvarial bone defects in SD rats. Micro-CT, real-time PCR, and histological analyses with H&E and Masson’s trichrome staining revealed the effective bone regeneration on the scaffolds using HA-PLGA/PEG/BMP-2 blend systems. |
