Sequence analysis and functional characterization of downstream region of the Clavulanic Acid Gene Cluster from Streptomyces clavuligerus |

Clavulanic acid (CA) is an important antibiotic produced by Streptomyces clavuligerus. Clavulanic acid is a potent inhibitor of β-lactamase enzymes and is of demonstrated value in the treatment of infections by β-lactam-resistant bacteria. Here, we report the sequencing of 12kb gene cluster from the downstream region revealing eight putative ORFs (from clav3 to clv10). We inactivate the clv6 gene and disrupted mutant strains were confirmed by southern blot experiment. And the productions of clavulanic acid of these two kinds of strains were analyzed by HPLC, and the yield of clavulanic acid in the clv6 mutants had a remarkably lower production. |
