Proteases play important roles in various cellular processes and thus abnormal activities of them are related with diseases. Methods have been developed to measure activities of these enzymes, most of which, however, are based on sophisticated instruments or complicated procedures. Needs for simple and reliable methods are increasing nowadays in particular for monitoring and management of patients. In this presentation, a POCT method is described to measure the activities of proteases, which is based on the pregnancy test strip detecting human chorionic gonadotropin (hCG). The hCG protein was engineered to interact with solid materials, and the artificial interaction can be prohibited by the activity of a protease of interest. Proteolytic cleavage by the enzyme is translated into free hCG in solution that can be detected by a commercial pregnancy test strip. The platform technology provides a simple and sensitive POCT method for disease-related proteases. |
