Regulation of Endocytosis Pathway of Polyplexes by Osmotic Polymeric Gene Carriers |

Degradable poly(amino ester)(PAE)s have received significant attention owing to their potentials in vitro and in vivo success of gene therapy. Amongst different PAEs, our PAE carrier composed of low molecular weight polyethylenimine (PEI) and PEGDA showed excellent DNA and siRNA delivery efficiency in vitro and in vivo. PAE-mediated Akt1 shRNA delivery efficiently decreased lung cancer cell survival, proliferation, and metastasis. Furthermore, folate conjugated PAE showed marked anti-tumor targeting potential of receptor-mediated endocytosis with low toxicity during and after therapy using TAM therapeutic gene. Recently, our PAE carriers composed of osmolytes and PEI have been designed for efficient gene transfection through regulation of shifting into cavolar endocytosis by hyperosmotic polyol part in the polymers and thus facilitated evasion of lysosomal degradation. These results suggest potential application of multi-functional PAEs for therapeutic DNA and siRNA delivery. |
