제목 Focal adhesive nano-pillar array for differentiation of human mesenchymal stem cell (hMSC)
초록 In order to differentiate hMSCs according to the number of focal adhesion points, we prepared nano-scale pillar arrays with different pillar-to-pillar distances. The silicon wafer template with nano-hole arrays was fabricated by e-beam lithography. Nano-pillar array was antitypically prepared using poly (dimethylsiloxane) (PDMS) against the master. The nano-pillar array was successfully fabricated at spacing of 3, 5, 10 μm with diameter of 500nm and height of 1μm on PDMS replica. The expression of focal adhesion proteins of hMSC cultivated on the patterned PDMS were significantly increased at the end of filopodias contacted with the top of pillar. A broad gene profiling of hMSC on the patterned PDMS was analyzed by Microarray. Genes related to neuronal differentiation, adhesion molecule signaling and cell cycle were regulated by nano-pillar array, suggesting that the induction of neuronal differentiation was associated with alteration of ECM signaling or cytoskeleton arrangement.
