Neuro-differentiation of Bone marrow Stromal Cells using β-mercaptoethanol |

In this study, we induced neuro-differentiation in the rat BMSCs by β-mercaptoethanol (BME). For isolation of rat BMSCs, females were dissected from Fisher rat (4-week of age). Here, BMSCs were expanded as undifferentiated cells in culture for more 4 passages. BMSCs were maintained in DMEM/10% FBS for 2 days. To initiate neural differentiation, the media (DMEM/10%FBS) were plated with BME of different concentration (Long-term cultures). Morphological change of BMSCs was exhibited at 1h to 6days. To induce neural differentiation of BMSCs in long-term cultures, differentiation viability of BMSCs was superior lower BME concentration than high concentration. But, very high and low concentration were not induced. After 12h, most of the BMSCs displayed distinct neuronal shape at 1mM BME concentration. Some cells contacted with each other through their branches. This research was supported by KMOHW (0405-BO01-0204-0006) and SCRC (SC3100). |
