Suppression of initial burst in PLGA Microspheres using polymeric complex |

ET (Escitalopram) encapsulated PLGA microspheres can be released for a long time, but industrial application is difficult due to initial burst. The goal of this study is to complex ET with pGA to sustained release. ET-pGA was prepared by ionic interaction between escitalopram oxalate and poly-galactronic acid. This was encapsulated in a PLGA microspheres by an s/o/w solvent evaporation method. As a result of analysis by UV-spectroscope and SEM microscope, the loading efficiency of microspheres was about 20% and the encapsulation efficiency was 85%. It was confirmed that ET-pGA-loaded microspheres were reduced in initial burst relative to the control group. The reason is that the drug-polymer chain was delayed in dissolution time in PBS. These properties are expected to lead to a stable sustained release of the drug in the polymeric complex form of ET-pGA. |
