제목 Engineered 3D Skeletal Muscle for Identifying Therapeutic Myokines
초록 A growing amount of evidence classifies skeletal muscle as an endocrine organ that can release secretory factors, called myokines. Despite the wealth of studies reporting beneficial effects of these putative factors, the exact mechanistic actions of myokines remain elusive due to technical challenges in the identification. Thus, we engineered a 3D microphysiological system that recapitulates native properties of contracting myofibers and integrated it with a biorthogonal proteome labeling system that can detect newly synthesized proteins. Our 3D muscle construct not only enables precise identification of bona fide myokines but also puts us one step closer to developing myokine-based therapy for late-onset lifestyle disorders or for patients in need of exercise therapy but cannot perform physical activities.
