제목 Cooperative binding of c-reactive protein to molecularly imprinted phosphocholine monolayer
초록 We report molecular imprinting of c-reactive protein(CRP) on a gold surface and the binding characteristics of the imprinted monolayer(MIM). 12-Mercaptododecyl phosphocholine (PC12SH) was synthesized and five molecules of PC12SH were reacted with one CRP molecule. The CRP-PC12SH complex was allowed to adsorb onto a gold surface, preserving the ligand arrangement of pentameric CRP-specific binding sites. The blocking was performed by BSA or 2-mercaproethanol(ME). The KA was calculated by Langmuir isothermal equation, using SPR measurement. In case of BSA-blocked substrate, the KA of MIM(2.65X109/M) was approximately 8.6 folds higher than that of non-imprinted monolayer(NIM). With ME-blocked substrate, the KA of MIM was obtained as 6.08X107/M. In Hill polt analysis, the MIM showed non-homogeneous binding characteristics on dependent of CRP concentration, which showed increased cooperativity in CRP binding at high CRP concentration. However, NIM showed homogeneous binding characteristics.
