제목 Robust Grafting of Cell-derived Extracellular Matrix on A Stent for Outgrowth Endothelial Cell Delivery
초록 Current stents including bare metal stent (BMS) and drug-eluting stent (DES) have several disadvantages for reendothelialization. The former has a poor surface to make endothelial progenitor cells (EPCs) adhere and the latter has a deteriorative effect on the viability of EPCs. To promote reendothelialization, we focused on a direct delivery of outgrowth endothelial cells (OECs), using an extracellular matrix (ECM)-coated cobalt-chromium alloy (CoCr) stent. For tight immobilization of an intact ECM on CoCr stent, we pre-coated polydopamine (pDA) and fibronectin (FN) on CoCr stent. The only CoCr-pDA-FN layer had the intact ECM layer on the surface after decellularization, compared to bare CoCr and CoCr-pDA layer. In addition, the stably immobilized ECM layer on CoCr-pDA-FN had higher efficacy than BMS in terms of the adhesion and growth of OECs. Thus, the ECM-immobilized stent would be a useful tool to deliver angiogenic cells into a damaged artery for rapid reendothelialization.
