Label free analysis of protein multivalent binding using bioresponsive hydrogels and SPR |

Identification of the interactions of a protein with other proteins or small molecules is vital for the understanding of biochemical pathways. Multivalent protein binding (MPB) plays a major role in regulating many biological functions but currently the determination of multivalent binding over monovalent binding requires complex processes and instruments. Here, we report a label-free, hydrogel-based SPR sensor for the quick determination of MPB and its kinetics. By conjugating MPB-capable moieties such as biocytin to the hydrogel modified surface of a SPR chip, we were able to evaluate MPB events. Using our method, we were able to observe enhanced signals from protein multivalent binding as compared to monovalent or non-specific binding. |
