제목 Induction of synergistic prostate cancer cell death via co-delivery of Bcl-xL shRNA and doxorubicin using aptamer conjugated smart polyplex
초록 A synergistic combination cancer therapy was investigated upon co-delivery of shRNA against Bcl-xL and doxorubicin (DOX) using aptamer conjugated smart polyplex (ASPs). Branched polyethyleneimine grafted with polyethylene glycol (PEI-PEG) was synthesized as a vehicle for shRNA delivery and its surface was further conjugated with anti-PSMA aptamer (APT) for selective delivery of ASPs to prostate cancer cells. ASPs were finally obtained after intercalation of DOX as a form of shRNA/PEI-PEG-APT/DOX conjugates. Cell viability test and FACS analysis of pEGFP expression against PC3 and LNCaP cells proved that the synthesized ASPs were highly biocompatible and had strong cell selectivity. Consequently, IC50 values of ASPs were approximately 17-fold less than that of the simple mixture of drug and gene. These results suggest that the co-delivery of anti-cancer drug and shRNA against Bcl-xL using synthethized ASPs may widen the therapeutic window and allow selective cancer cell killing.
