ED-B targeting peptide aptamer conjugated SPION (BPBEDB-SPIONs) for brain tumor MR imaging |

Here, we developed a novel EDB-targeted peptide aptamer conjugated Superparamagnetic iron oxide nanoparticles (BPBEDB-SPIONs) for brain tumor diagnosis. Our research group found EDB targeting peptide aptamer. In this research, we designed amphiphilic polymer coated SPION which contained EDB targeted peptide aptamer conjugated DSPE-PEG2000,DSPE-PEG2000-Methoxy and Rhodamine-DSPE via nanoemulsion method. Comparing with non-targeting SPION,BPBEDB-SPIONs showed higher uptake in EDB over-expressing U87MG cell line. In vivo experiments, EDB peptide conjugated SPION was accumulated more at tumor site compared to non-targeting SPION in U87MG tumor bearing mice. MRI signal at U87MG tumor site of BPBEDB-SPIONs group was lower, indicating the specific targeting and uptake of BPBEDB-SPIONs. Furthermore, we confirmed that the BPBEDB-SPIONs was accumulated only at EDB expression site. These results suggests that our BPBEDB-SPIONs has high potential as MR imaging of EDB over-expressing tumor. |
