제목 Bone Regeneration through pDNA Complex-Loaded Asymmetrically Porous GBR Membrane Using a Mini-Pig Model
초록 Recently, bone regeneration using tissue engineering technique combined with a local gene delivery system has been considered as one of the promising therapeutic methods. In this study, we prepared plasmid DNA (with BMP-2 encoding) complex-loaded polycaprolactone (PCL) asymmetrically porous membrane for enhanced bone regeneration. For effective plasmid DNA release from the membrane and the following transfection of plasmid DNA into stem cells, the PCL membrane was hydrophilized with Pluronic F127 and the plasmid DNA was complexed with polyethyleneimine-polyethylene glycol (PEI-PEG). The pDNA/PEI-PEG complex was continuously released from the PCL/Pluronic F127 membrane for more than 3 months. The bone regeneration behavior of the pDNA complex-loaded PCL/Pluronic F127 membrane were investigated using a miniature pig model. The pDNA complex-loaded PCL/F127 membranes showed faster bone regeneration than the control (blank) and PCL/Pluronic F127 membrane without plasmid DNA.
