Controlling mRNA secondary structure for efficient gene expression |

Efficient expression of the heterologous genes in lab-friendly microorganisms is important for endowing host cells with new biological properties. It is well known that mRNA secondary structures in the non-coding regions directly influence the protein synthesis by altering ribosome accessibility and mRNA stability. We have investigated how the mRNA secondary structures in the 5’ proximal coding region alter the protein synthesis using Escherichia coli and Saccharomyces cerevisiae as expression hosts. We found that strong secondary structures in the 5’ proximal coding region retarded protein expression in both prokaryotic and eukaryotic systems, indicating that synthetic gene design should include the consideration of secondary structure stability in the 5’ proximal coding region as well as the codon bias in the whole DNA sequence. |
