제목 Metabolic Engineering of 3-Hydroxypropionic Acid-producing Escherichia coli  via Malonyl-CoA Dependent Pathway
초록 3-Hydroxypropioic acid (3-HP) is a promising platform chemical for the production of a variety of industrial compounds. It can be biologically produced from the reduction of malonyl-CoA catalyzed by malonyl-CoA reductase (MCR). Although this malonyl-CoA pathway is highly attractive to produce 3-HP, it has been known that the activity of this pathway is not sufficient. Moreover, it requires a precise flux optimization as malonyl-CoA is also an important precursor for fatty acid synthesis. In this study, we engineered Escherichia coli to achieve high 3-HP production. At first, we amplified the MCR pathway by maximal expression of genes encoding MCR enzyme as well as other pathway enzymes under strong synthetic UTRs and promoters. Thereafter, for flux optimization, the expression of the first gene in fatty acid synthesis from malonyl-CoA, was varied under different-strength promoters. Our engineering strategy improved the 3-HP titer by 7.00-fold increase compared to the parental strain.
