We are reporting that molecular weight of the PEG as well as the molecular weight of the polypeptide is critically important in determining the secondary structure of a polypeptide. As a model system, we synthesized the poly(ethylene glycol)-polyalanine, where the molecular weight of PEG was varied over 1,000 ~ 5,000 daltons at a fixed molecular weight of PA at 700 daltons. In addition, we varied the molecular weight of PA over 700 ~ 1,500 daltons at a fixed molecular weight of PEG at 2,000 daltons. The secondary structure of the polypeptides was investigated by circular dichroism spectroscopy and FTIR spectroscopy. Due to the amphiphilic nature of the PEGylated polypeptide, they tend to self-assemble in water. |
