The extremophilic bacterium Deinococcus radiodurans which has outstanding tolerance to extreme stress conditions has been considered as a fascinating microorganism for industrial applications. However, this bacterium has shortcomings such as non-replicable plasmids and the lack of tools for saving antibiotic resistance gene of the genome and repetitive genetic manipulation. Although constant exertions have been occurred to develop tools, with regard to industrial applications, more efficient tools for multiple knockout of D. radiodurans strain has not been developed yet. Therefore, we designed a multiple knockout method based on Cre-lox system for metabolic engineering D. radiodurans R1 and successfully deleted the dr0053 gene of the strain. This result shows it will make the strain applicable microorganism to industry by engineering D. radiodurans. |
