Biodegradable coating and local delivery of tauroursodeoxycholic acid for coronary stent application |

Coronary artery diseases are usually treated by stent implantation. Currently used bare metal stent (BMS) and drug-eluting stent (DES) showed an excellent clinical efficacy. However, over a long-term follow-up, BMS and DES causes restenosis due to the vascular smooth muscle cell (VSMC) hyperplasia and late-thrombosis caused by delayed re-endothelialization due to drug toxicity, respectively. Tauroursodeoxycholic acid (TUDCA) has been reported to have an effect on inhibiting neointimal hyperplasia by up-regulating apoptosis of VSMCs. In addition, TUDCA promotes proliferation and migration of endothelial adult and progenitor cells. In this study, we utilized TUDCA as an alternative drug of currently commercialized drugs such as sirolimus. With poly lactic-co-glycolic acid (PLGA) coating, the drug could be locally delivered to the stented site with a steady release. To the best of our knowledge, it can be a promising way to promote blood vessel recovery without side effects of drug. |
