A New Class of Bio-Inspired In Situ-Forming Long-lasting Themo-Sensitive Injectable HA/Pluronic Hydrogels Using Dopamine-Thiol Reaction |

Hydrogels based on hyaluronic acid (HA) are widely studied as biocompatible and biodegradable materials for cartilage regeneration, wound healing and post-surgical anti-adhesion. However, poor mechanical properties exclude many biomedical applications. Although many approaches aimed to reduce the problems, synthesis of rapid in situ-gelling injectable HA-based hydrogels is still challenging. Thermo-gelling injectable hydrogels based on poly(ethylene oxide)-poly(propylene oxide)-poly(ethylene oxide) (PEO-PPO-PEO) have been used for injectable cell therapy, tissue regeneration, protein delivery and wrinkle filler applications. Here, we synthesized HA-dopamine conjugate (HA-DN) and thiol-terminated Pluronic (Plu-SH) and developed a bio-inspired in situ-forming thermo-sentivie long-lasting injectable hydrogel by combining HA-DN and Plu-SH using dopamine-thiol reaction for rapid and robust crosslinking under physiological conditions. |
