제목 Oral siRNA delivery to CD44 receptor target hepatic cancer therapy using the enterohepatic circulation pathway
초록 Recent studies on siRNA therapy showed promising results in treating Cancer. However, obstacles including enzymatic degradation, lack of targeting specificity and toxicity restricted their clinical applications. Herein, we developed a Hyaluronic acid-Taurocholic acid (HA-TCA) coated oral gene delivery system to treat hepatic cancer. HA, a natural polysaccharide, can actively target the CD44 receptors expressed over cancer cells. TCA is a kind of bile acid that play a critical role in enterohepatic circulation. In this study, HA-TCA was synthesized and wrapped over protamine/siRNA nano-complex to treat hepatic cancer. Size, zeta potential and gel electrophoresis studies confirmed the formation of stable siRNA loaded nanoparticles. Further, in vitro studies confirmed the siRNA activity with minimal toxicity in MDCK cell lines. In conclusion, we believe that HA-TCA capped siRNA nanoparticles could endure the GI degradations and aid in CD44 mediated targeted hepatic cancer therapy.
