To diminish overused antibiotics, rapid and accurate diagnostic assay for discerning Gram-positive bacteria should be developed. Here, we describe colorimetric assay based on the vancomycin conjugated nanobeads aggregation. The vancomycin conjugated nanobeads can induce the agglomeration with Gram-positive bacteria via hydrogen bonding between vancomycin and the subunits on the Gram-positive bacterial cell wall. The agglomeration can be easily detected by naked eye. Also, we demonstrate the porous filter system introduced to the colorimetric assay. By applying the colorimetric assay to various bacterial species, we confirmed that the nanoparticle agglomeration based colorimetric assay is rapid, simple, and accurate for detection of Gram-positive bacteria. This colorimetric assay can be utilized as a novel Gram-positive bacterial diagnostic tool in point-of-care settings. |
