The use of low molecular weight fluorochromes for in vivo imaging is limited by light attenuation through biological tissues and by the behavior of low molecular fluorochromes in vivo. To overcome these limitations, the modular nanoprobes (PN) were designed to allow independent variation of functional groups. We have attached TO-PRO 1 to Feraheme (FH) NP’s. The DNA binding fluorochrome TO-PRO 1 (TO) was attached to FH, yielding a NP functionalized with multiple TO’s. TO-FH failed to bind normal cells, but treatment with chemotherapeutic agents or detergents yielded necrotic cells that bound TO-FH. PEGylated Nanoprobes (PN’s) are passively targeted nanomaterials featuring PEG-controlled pharmacokinetics, detectability by fluorescence/SPECT or fluorescence/PET. We describe the reaction between an NHS-ester of rhodamine (Rho) and clinical protamine (Pro), which yields RhoPro, and show that the RhoPro produced has a photodynamic activity, causing rapid light induced cell death. |
